High Multiplex Proteomics Using a Proximity Extension Assay with an Automated Liquid Handling System

Large-scale analysis of proteins is an integral part of the multi-omics approach in understanding human biology and identifying therapeutic targets. However, the full potential of proteomics has not been realized due to common limitations in technologies, including high sample consumption, low specificity, high inter- and intra-assay variations and narrow dynamic range. Here, the featured speakers discuss the combined benefits of a proximity extension assay with an automated liquid handling system.

The Olink proximity extension assay (PEA) technology combines the sensitivity of a polymerase chain reaction (PCR)-based method with the specificity of an antibody-based detection method and the scalability of next-generation sequencing (NGS). This provides a unique solution enabling simultaneous screening of thousands of proteins, using only a few microliters per sample. The PEA technology has a dynamic range of 10 orders of magnitude and requires no sample fractionation. Therefore, the Olink® Explore is an ideal platform for analyzing the proteomes of various sample types.

Eppendorf has partnered with Olink to automate the Olink® Explore workflow on the Eppendorf epMotion® 5075 liquid handler, enabling a simplified setup for a high-throughput screening assay. This is a flexible and user-friendly automated liquid handling platform that significantly reduces assay variation, improves data reproducibility and decreases hands-on time. With high pipetting accuracy and precision, this liquid handler provides a robust and highly precise automation solution.

Kanhav Khanna, Field Application Specialist, Eppendorf

Mr. Kanhav Khanna joined Eppendorf in 2016 and currently works as an Application Specialist for Eppendorf’s Automated Liquid Handling. He is responsible for performing training, and method development on Eppendorf’s epMotion® liquid handlers. He also coordinates the generation and production of technical literature such as training courses, user guides and manuals. Kanhav holds a Master of Science degree in Biology and Biotechnology from Texas Tech University, where he studied small RNA (sRNA) biogenesis. He has many years of experience with next-generation sequencing (NGS) including sample processing and quality analysis. Prior to joining Eppendorf, Kanhav worked as a Research Investigator in the sequencing core at MD Anderson Cancer Center (Houston, Texas, USA) responsible for assessing new NGS technologies and improving existing NGS workflows.

Daniel Frederick, Senior Field Application Scientist, Olink

Daniel Frederick, PhD is a Senior Field Applications Scientist for Olink Proteomics that joined the company in July 2020 to support external users and new core facilities in New England. Prior to that he was functioning in a similar role at the Fluidigm Corporation supporting all suspension mass cytometry applications and customers for the New England territory. His background is strongly grounded in immunology from his graduate work at Tulane University and postdoctoral work at the University of North Carolina, Chapel Hill. His combined experience in academia and the life sciences industry has given him considerable vision on both single-cell and liquid sample proteomics.
