Primary Duties
The Research Associate – AAV Capsid Engineering is an important member of the Discovery Research Team and participates in the development of Spark’s gene delivery platforms. She/he/them will be critical for the development of novel AAV capsids with enhanced properties, in particular the ability to transduce specific cell types. The discovery of such capsids will greatly facilitate the translation from in vitro through in vivo proof of concept and ultimately the clinic.
Responsibilities
- Assisting in the characterization and validation of novel AAV capsid libraries and the use of these libraries for selecting cell-type specific AAV variants and AAV variants with other improved properties. Characterizing lead candidates for preclinical work. This includes executing experiments at the bench and keeping up with relevant literature.
- Organizing, recording, and discussing experimental data.
- Participating in lab meetings and administration work.
- Cloning of plasmids used for the production of AAV capsid libraries and their characterization.
- Establishing and executing vector biodistribution studies.
- Establishing and executing transgene expression studies both in vitro and in vivo.
- Assisting in establishing approaches that allow the isolation of AAV capsid variants that transduce specific cell types.
- Collaborating with the Genomics and Data Science teams to determine the sequence space of AAV DNA libraries and AAV DNA recovered from desired tissues.
- Actively participating in joint programs and collaborations with other teams within the Spark Research & Technology division.
- Writing study reports and keeping detailed records of lab work.
The Research Associate position requires experience with molecular biology and/or virology:
- Expertise in the design and construction of AAV/bacterial/phage libraries highly desirable.
- Basic knowledge in NGS and bioinformatics (in collaboration with experts in these fields) to design improved second generation libraries.
Education and Experience Requirements
- Typically has a Bachelor’s degree in a relevant biological discipline and 0-3 years of related experience.
- Experience in general lab techniques such as molecular cloning, qPCR, cell culture, ELISA, plate-based luciferase and/or enzymatic activity assays, SDS-PAGE/Western blotting.
- Familiarity with AAV gene therapy is not required.

- Extensively collaborates with other members of the capsid engineering team.
- Interacts with and supports other members of the Research & Technology Department.
