In vivo CAR-T therapy requires durable, yet transient CAR expression coupled with efficient, T cell-specific delivery. This webinar will focus on how innovations in RNA design and next-generation lipid nanoparticle (LNP) technology are enabling direct in vivo engineering of T cells, replacing conventional ex vivo manufacturing. This webinar will explore key advances in CAR construct design and novel LNP formulations to improve the precision, potency and safety of in vivo CAR-T therapies.

Modular “drop-in” CAR plasmids that streamline both construct design and RNA production will be introduced. These vectors produce mRNAs optimized for CAR expression and support flexible swapping of antigen-binding domains, enabling rapid screening and efficient customization. This approach may be particularly useful for researchers who are new to CAR-T development.

The featured speaker will present recent innovations in RNA design to enhance expression, durability and tissue specificity. These innovations include a modified poly(A) tail that improves transcript stability and translational efficiency and a T cell-specific internal ribosome entry site (IRES) that, when incorporated into circular RNA (circRNA), restricts CAR expression to T cells and reduces off-target effects.

This session will also highlight novel LNP technology engineered for T cell-directed delivery, including exosome-like and spleen-tropic LNPs that enrich delivery to lymphoid tissues where T cells are concentrated and improve CAR expression. VHH site-specific conjugated LNPs that employ single-domain antibody fragments for precise T cell targeting will also be discussed. Collectively, these advances in CAR construct design, RNA engineering and targeted LNP systems represent a comprehensive toolbox for next-generation in vivo CAR-T therapy development.
